PREPARATION OF SELF-ASSEMBLED BOVINE SERUM ALBUMIN NANOPARTICLES LOADED WITH CURCUMIN AND INVESTIGATION OF FACTORS AFFECTING in vitro DRUG RELEASE
Main Article Content
Abstract
Curcumin is a natural bioactive compound with many important biological effects; however, its application is limited due to poor solubility, low stability, and low bioavailability. Self-assembled albumin nanoparticle systems can improve the solubility and bioavailability of curcumin without the need for crosslinking. In addition, the drug release process depends on factors such as pH, temperature, and reducing agents (glutathione). Therefore, the preparation of self-assembled albumin nanoparticles loaded with curcumin and the investigation of factors affecting drug release are essential. The research group successfully prepared self-assembled albumin nanoparticles loaded with curcumin, with a particle size of 156,84 nm, a PDI of 0,154 (<0,2), and a zeta potential of −30 mV. The results also showed an encapsulation efficiency of approximately 88% and a drug loading capacity of about 7% when using an albumin concentration of 20 mg/mL and a reducing agent concentration of 10 mM. Furthermore, the self-assembled nanoparticle system remained relatively stable for 7 days (particle size <250 nm, PDI <0,3) in distilled water and physiological buffer. The curcumin release profile was environment-dependent, with higher release observed at 37°C, under acidic conditions (pH 5,5), and in the presence of glutathione (GSH).
